KLH-conjugated phosphopeptide containing the pTEpY motif of Erk1 and Erk2.
Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested. Recommended Dilution: Western Blot: 0.1-1ug/ml detects pMAPK in RIPA lysates from EGF-stimulated human A431 carcinoma cells. Non-stimulated A431 and EGF-stimulated A431 cell lysates were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-pMAPK (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: Use 1-10ug per immunoprecipitation. Optimal dilutions to be determined by the researcher. Positive Antigen Control: EGF-stimulated A431 lysate. Use 20ug per lane for minigels. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
