Immunogen: HPLC purified murine recombinant MAP Kinase.
ERK2 is activated by MEK2, which phosphorylates neighboring threonine 183 and tyrosine 185 residues. ERK2 is a serine/threonine kinase that phosphorylates MAP2 and myelin basic protein. This kinase is an important proximal component of the MAP kinase pathway involved in transeitting the signals from growth factors, neurotransmitters and hormones at the cell surface to the transcriptional events in the nucleus. Applications: Suitable for use in FLISA, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilutions: Immunoprecipitation: 4ug immunoprecipitates MAP Kinase from a RIPA lysate from mouse 3T3 cells. Boil the cell lysate and let cool before adding the antibody as this antibody only binds to denatured MAP Kinase. Western Blot: 0.5-2ug/ml detects MAP Kinase in RIPA lysates from human A431 carcinoma cells. A non-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with M2362 (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody (HRP) and a chemiluminescence detection system. A previous lot detected MAP Kinase in mouse 3T3 fibroblasts and rat L6 cells. Optimal dilutions to be determined by the researcher. Included Positive Control: A0001-50: Non-stimulated A431 cell lysate. Add 2.5ul of 2-mercaptoethanol per 100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
