p38 MAP kinase (MAPK), also called RK or CSBP, is the mammalian orthologue of the yeast HOG kinase which participates in a signaling cascade controlling cellular responses to cytokines and stress. Four isoforms of p38 MAP kinase, p38a, b and g (also known as ERK6 or SAPK3) and delta (also known as SAPK4) have been identified. Similar to the SAPK/JNK pathway, p38 MAP kinase is activated by a variety of cellular stresses including osmotic shock, inflammatory cytokines, lipopolysaccharides LPS), UV light and growth factors. MKK3, MKK6 and SEK activate p38 MAP kinase by phosphorylation at Thr180 and Tyr182. Activated p38 MAP kinase has been shown to phosphorylate and activate MAPKAP kinase 2 (3) and to phosphorylate the transcription factors ATF-2, Max and MEF2. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunofluorescence. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000. Iincubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20. Immunohistochemistry (Paraffin): 1:50. Antigen retrieval: heating in citrate buffer. Dilute antibody in TBS-Tween plus 5% normal goat serum. Immunofluorescence (IF-IC): 1:100 Optimal dilution determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
