MaxLight(TM)490 is a new Blue-Green photostable dye conjugate comparable to DyLight(TM)488, Alexa Fluor(TM)488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm), Emission (515nm), Extinction Coefficient 73,000. Integrins are heterodimeric cell surface receptors composed of alpha and beta subunits, which mediate cell-cell and cell-extracellular matrix attachments. Aberrant integrin expression has been found in many epithelial tumors. Changes in integrin expression have been shown to be important for the growth and early metastatic capacity of melanoma cells. Integrin alpha-v beta-6 is upregulated in cancers and during tissue remodelling but is absent from resting adult tissues. Integrin alpha-v beta-6 promotes invasion and correlates with poor survival and therefore makes a promising therapeutic target. Applications: Suitable for use in Immunohistochemistry, Flow Cytometry and Immunocytochemistry. Not recommended for Western Blot or Immunoprecipitation. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry (Acetone fixation): Effective for human tissue, especially in circumstances where up-regulation of the receptor is being evaluated. Not suitable for use on formalin-fixed or paraffin-embedded tissues. May not detect the low-level basal expression of Integrin alphaV, beta6 in epithelia of healthy adults. Immunocytochemistry: Effective for F6, HT-29, H441 cell lines. (Other cell lines not tested). Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight(TM)490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
