Integrins are a family of dimeric proteins that mediate cell-cell and extracellular matrix adhesion. Each VLA subtype (VLA-1, -2, -3, -4 and -5) is composed of a distinct alpha subunit noncovalently associated with a common beta subunit, as an alpha-beta dimer. Integrin VLA-3 (very late antigen, alpha 3/beta 1) is a cell surface receptor that binds to fibronectin, lamin, collagen type I and VI. VLA-3 is typically restricted in normal tissue but is widely expressed in many cultured tumor cells and correlates with the degree of invasiveness of malignant melanoma cells. Applications: Suitable for use in Immunohistochemistry, ELISA, RIA, Immunoprecipitation and Immunocytochemistry. Other applications have not been tested. Recommended Dilution: Immunohistochemistry: Acetone fixed, paraffin embedded sections Immunoprecipitation: Under non-reducing conditions, two chains with apparent molecular weight of 110 and 150kD. In the presence of a reducing agent (2% B-ME), a single molecular species of 130kD is detected. Under reducing conditions both alpha3 and beta1 subunits run with the same apparent molecular weight. Optimal dilutions to be determined by the researcher. Positive Controls: Cell Line: KJ29 renal carcinoma adherent cells Tissue: Kidney. Reacts with basal layer podocytes and basement membrane of distal tubules. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
