Granzyme A is a member of the granzyme family. Granzyme A is the most abundant serine protease in cytotoxic T lymphocytes (CTL) and natural killer (NK) cells. Granzyme A has a specific function in CTL and NK cells. It induces caspase-independent cell death when introduced into target cells by perforin. Human granzyme A is synthesized as a precursor (262 residues) with a signal peptide (residues 1-26), a propeptide (residues 27-28) and a mature chain (residues 29-262). Activity: Measured by its ability to cleave a colorimetric peptide substrate, N-carbobenzyloxy-Gly-Arg-ThioBenzyl ester (Z-GR-SBzl), in the presence of 5,5Dithio-bis (2-nitrobenzoic acid) (DTNB). Endotoxin: 1.0 EU per 1ug of the protein by the LAL method. Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at -20C. Reconstitute at 100ug/ml in sterile 25 mM HEPES, 150mM NaCl and 10mM CaCl2, pH 7.5. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
