For the quantitative determination of rat thyrotropin-releasing hormone in serum, plasma, and tissue homogenates. Sensitivity: 0.25pg/ml Detection Range: 0.5-100pg/ml Specificity: This assay has high sensitivity and excellent specificity for detection of rat TRH. No significant cross-reactivity or interference between rat TRH and analogues was observed. Assay Principle: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with goat-anti-rabbit antibody. Standards or samples are added to the appropriate microtiter plate wells with an antibody specific for TRH and Horseradish Peroxidase (HRP) conjugated TRH. The competitive inhibition reaction is launched between with HRP labeled TRH and unlabeled TRH with the antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of TRH in the sample. The color development is stopped and the intensity of the color is measured. Kit Components: Microtiter Strips, 1x96 wells Standard, 6x0.5ml Antibody, 1x6ml HRP-conjugate, 1x6ml Wash Buffer (20X), 1x15ml Substrate A, 1x7ml Substrate B, 1x7ml Stop Solution, 1x7ml Storage and Stability: Store all kit components at 4C. Stable for 6 months after receipt. Once opened, the kit is stable for 1 month providing this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
