HDAC6-IN-71 (Compound 24) is an HDAC6 inhibitor with IC50 values of 13.68 nM for HDAC6 and 443.12 nM for HDAC1. It effectively suppresses NO production in mouse macrophages with an IC50 of 2.31 µM. By inhibiting the HDAC6-NF-kappaB signaling pathway, HDAC6-IN-71 reduces the phosphorylation levels of IkappaB-alpha and IKK-alpha/beta, and inhibits the expression of downstream inflammatory proteins COX-2 and iNOS. Additionally, it significantly alleviates ulcerative colitis in mice.
