Specificity of the antibody was verified by its ability to immunoprecipitate a5b1 heterodimers from 125-I-surface labeled cells, reciprocal depletion of VLA-5 antigen from cell lysate with antiserum against the cytoplasmic domain of a5 integrin subunit, and immunoprecipitation of VLA-5 integrin from cells known to express this integrin. Cell lines tested to express a5b1, include: P19 cells, mouse melanoma B16F1 cells, mouse squamous carcoma SCC cells and thymocytes. Characterization by immunoprecipitation was typically analyzed by SDS-PAGE using 6% gels under non-reducing conditions. Applications: Suitable for use in the Inhibition of cell adhesion to fibronectin, Immunoprecipitation, Flow Cytometry and Western Blot. Other applications not tested. Recommended Dilutions: Inhibition of Cell Adhesion to Fibronectin: 10ul/ml Note: where cell adhesion to fibronectin involving other fibronectin receptors, such as VLA-4 (a4b1) integrin, is involved, inhibition of cell adhesion by BMC5 alone may not be complete. Immunoprecipitation from lysate of 10e6 cell equivalent: 2ul Flow Cytometry: 10ul/ml Western Blot: Non-denaturing gel Optimal dilution determined by the researcher. Hybridoma: Fusion of spleen cells from a Fisher rat immunized with mouse embryonal carcinoma P19 cells with myeloma P3X63Ag9.653 cells Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
