Gai2 is a 40-41kD alpha-subunit of heterotrimeric protein that shares sequence homology with Gai3 and Gai1. All these G proteins can be ADP-ribosylated at the carboxy terminal Cys of the CAAX sequence, interfering with receptor coupling. Gai2 is expressed widely in tissues and cell lines. Activation of PLC-b by Gai-coupled receptors is the result of release of Gbg subunits from Gai. Gai2 regulates potassium channel activation. Mutation at Arg179 has been identified in tumors. This and the Q205L mutant inhibit adenylyl cyclase. Applications: Suitable for use in FLISA and Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
