Verwendungszweck

Der SYPHILIS RPR TEST ist ein nicht-treponemaler Flockungstest zum qualitativen und semiquantitativen Nachweis von Syphilis-assoziierten Reagin-Antikörpern in humanem Serum oder Plasma.

Der Test ist nur für den professionellen Gebrauch bestimmt und sollte nur von geschultem Personal durchgeführt werden.

Prinzip

Das im Kit verwendete RPR-Antigen ist eine Modifikation des VDRL-Antigens, das Kohlemikropartikel enthält, um leichter zwischen positiven und negativen Ergebnissen unterscheiden zu können. Enthält ein Serum Anti-Reagin-Antikörper, kommt es zur Ausflockung von Antigen-Kohlepartikeln. Im Fall von nicht reaktiven Proben bleibt die Probe-Antigen-Suspension homogen.

Funktionsweise

Der SYPHILIS RPR TEST wurde als Rapid-Plasma-Reagin-Test entwickelt, ein Screening-Test zum Nachweis von Reagin-Antikörpern, die auf eine Syphilis-Infektion hinweisen. Der Test basiert auf der Reaktion von Anti-Reagin-Antikörpern, die an Aktivkohle-Cardiolipin-Partikel binden und so sichtbare Agglutinate bilden. Der Test kann entweder qualitativ oder semiquantitativ durchgeführt werden. Im letzteren Fall werden positive Proben seriell mit einer physiologischen Salzpufferlösung verdünnt. Der Titer wird als die Endverdünnung angegeben, bei der ein positives Ergebnis erzielt wird.

Der Test umfasst eine RPR-Antigensuspension, bestehend aus Cardiolipin- und Aktivkohlepartikeln, sowie positive und negative Kontrollen.

Der Test wird auf einem Objektträger mit 10 Feldern durchgeführt. Objektträger, RPR-Antigen, Dispensernadel und Dispenserflasche sind im Lieferumfang des Tests enthalten.

Principle

The RPR antigen used in the kit is a modification of the VDRL antigen that contains carbon microparticles to facilitate differentiation between positive and negative results. If a serum contains anti-reagin antibodies, flocculation of the antigen-carbon particles occurs. In the case of non-reactive samples, the sample-antigen suspension remains homogeneous.

How It Works

The SYPHILIS RPR TEST was developed as a Rapid Plasma Reagin (RPR) test, a screening test for the detection of reagin antibodies that indicate a syphilis infection. The test is based on the reaction of anti-reagin antibodies, which bind to activated charcoal-cardiolipin particles and thus form visible agglutinates. The test can be performed either qualitatively or semiquantitatively. In the latter case, positive samples are serially diluted with a physiological saline buffer solution. The titer is reported as the final dilution at which a positive result is obtained.

The test includes an RPR antigen suspension consisting of cardiolipin and activated charcoal particles, as well as positive and negative controls.

The test is performed on a 10-well slide. The slide, RPR antigen, dispenser needle, and dispenser bottle are included with the test kit.
