Background

The protein encoded by this gene is a member of the G-protein-coupled receptor family. This protein is a receptor for interleukin 8 (IL8). It binds to IL8 with high affinity, and transduces the signal through a G-protein activated second messenger system. This receptor also binds to chemokine (C-X-C motif) ligand 1 (CXCL1/MGSA), a protein with melanoma growth stimulating activity, and has been shown to be a major component required for serum-dependent melanoma cell growth. This receptor mediates neutrophil migration to sites of inflammation. The angiogenic effects of IL8 in intestinal microvascular endothelial cells are found to be mediated by this receptor. Knockout studies in mice suggested that this receptor controls the positioning of oligodendrocyte precursors in developing spinal cord by arresting their migration. This gene, IL8RA, a gene encoding another high affinity IL8 receptor, as well as IL8RBP, a pseudogene of IL8RB, form a gene cluster in a region mapped to chromosome 2q33-q36. CXCR2 antibody [HL2604] detects CXCR2 protein at cell membrane by immunofluorescent analysis. Sample: THP-1 cells were fixed in ice-cold MeOH for 5 min. Green: CXCR2 stained by CXCR2 antibody [HL2604] (GTX639056) diluted at 1:200. Blue: Fluoroshield with DAPI (GTX30920).
Unboiled THP-1 whole cell and membrane extracts (30 μg) were separated by 10% SDS-PAGE, and the membranes were blotted with CXCR2 antibody [HL2604] (GTX639056) diluted at 1:1000 and competitor's antibody (Clone 48311) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.
CXCR2 antibody [HL2604] (GTX639056) detects CXCR2 protein by flow cytometry analysis. Sample: Non-transfected and transfected 293T cells were fixed in 4% paraformaldehyde at 4ºC for 15 min and washed with 0.1% saponin buffer to permeabilize the cell membrane. The cells were following stained with CXCR2 antibody [HL2604] (GTX639056) diluted at 1:50 or a Rabbit IgG isotype control (GTX35035) control at 4ºC for 1 hour.
Unboiled non-transfected (–) and transfected (+) 293T whole cell extracts (30 μg) were separated by 10% SDS-PAGE, and the membrane was blotted with CXCR2 antibody [HL2604] (GTX639056) diluted at 1:5000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.
